This book illustrates the importance of solid-state properties of pharmaceutical materials in the development of pharmaceutical products, and lays out general strategies for the physical characterization of solids using various analytical tools. First it covers essential knowledge on the various forms that solids can assume, including polymorphs, solvates/hydrates, salts, co-crystals, amorphous forms, crystal mesophases, and nanocrystals, as well as issues related to their relative stability and tendencies to undergo transformation. Secondly it presents the key methods of solid state analysis such as X-ray crystallography, X-ray powder diffraction, thermal analysis, microscopy, vibrational spectroscopy, and solid state NMR. Then it reviews critical physical attributes of pharmaceutical materials including particle size/surface area, hygroscopicity, mechanical properties, solubility, and physical and chemical stability, for small molecules and large molecules, such as proteins. Finally, it features applications of physical characterization in drug solid form selection, pharmaceutical analysis, product development, and manufacturing control using Quality by Design.
